This is mainly because large plasmid DNA has a large molecular weight, often over 100kb, and generally low copy number and low yield, and the efficiency and yield of adsorption using conventional centrifugal column adsorption membranes is very low and the permeation of membranes will interrupt these large molecular weight plasmid DNA.
